ABSTRACT

ObjectiveTo explore the effects of Baihu Jia Renshen Tang (BHRS) on the related molecules on the phosphatidylinositol-3-kinase/protein kinase B(PI3K/Akt)signaling pathway in the liver of MKR diabetic model mice. MethodThirty 6-week-old MKR mice were selected and fed on a high-fat diet for four weeks,followed by intraperitoneal injection of streptozotocin(STZ)for the diabetes model establishment. The model was properly induced in the case of the fasting blood glucose (FBG) of ≥11.1 mmol·L-1. After modeling,the mice were randomly divided into a model group,a BHRS group (12.09 g·kg-1·d-1),and a metformin group (0.065 g·kg-1·d-1),with 10 mice in each group. Ten FVB mice were assigned to the control group. The mice in the groups with drug intervention were continuously administered correspondingly for 28 days. After administration,the mice were sacrificed,followed by the oral glucose tolerance test (OGTT) and FBG detection. Serum very low-density lipoprotein(VLDL)content was determined by semi-quantitative enzyme-linked immunosorbent assay (ELISA). Four indexes related to blood lipid were determined by the biochemistry analyzer. Liver tissues were subjected to pathological examination by hematoxylin-eosin(HE)staining. Western blot was used to detect the protein expression of PI3K,Akt,phosphorylated(p)-PI3K,p-Akt,forkhead box protein O1 (FoxO1),insulin receptor(InsR),and insulin receptor substrate-2(IRS-2) in liver tissues of mice. Real-time polymerase chain reaction(Real-time PCR) was used to detect the mRNA expression of PI3K,Akt,FoxO1,InsR,and IRS-2 in liver tissues of mice. ResultCompared with the control group,the model group showed poor general conditions,abnormal glucose tolerance (P<0.05),increased FBG (P<0.01),abnormal blood lipid metabolism,increased serum total cholesterol (TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and VLDL (P<0.05),decreased level of high-density lipoprotein cholesterol(HDL-C)(P<0.05),fatty degeneration and obvious pathological changes of liver cells,reduced protein expression of PI3K,Akt,p-PI3K/PI3K,p-Akt/Akt,IRS-2,and InsR in liver tissues(P<0.05),increased protein expression of FoxO1(P<0.05),decreased mRNA expression of PI3K,Akt,IRS-2,and InsR in liver tissues (P<0.05),and increased FoxO1 mRNA expression(P<0.05). Compared with the model group,the BHRS group showed improved general conditions and glucose and lipid metabolism (P<0.05),improved pathological state of liver cells,increased protein expression of PI3K,Akt,p-PI3K/PI3K,p-Akt/Akt,IRS-2,and InsR in liver tissues(P<0.05),decreased protein expression of FoxO1(P<0.05),increased mRNA expression of PI3K,Akt,IRS-2,and InsR in liver tissues (P<0.05),and reduced FoxO1 mRNA expression(P<0.05). ConclusionBHRS can effectively reduce blood glucose,regulate blood lipid metabolism,and improve the pathological state of the liver in MKR diabetic mice,and its mechanism of action may be related to the regulation of the activity of molecules on the PI3K/Akt signaling pathway.

ABSTRACT

Autologous fat grafting (AFG) is a new postoperative plastic technique for breast cancer. The safety of this technique in oncology is a controversial issue in academic circles. In some basic experiments, it was found that adipocytes could promote the proliferation of surrounding cancer cells through autocrine and paracrine. However, clinical trial data show that this procedure does not increase the risk of postoperative recurrence or metastasis of breast cancer. Nowadays, there are variety of researches on the safety of this technique, but many limitations are found in these trials. Therefore, to accurately assess the relevance of these studies to any realistic clinical risk, it is necessary to include large sample of clinical studies. In this paper, the data of basic experiments and clinical trials in recent ten years were collected, and the safety of oncology was further analyzed and discussed, which will provide reference for clinical decision-making of postoperative breast reconstruction.

ABSTRACT

Objective To quantitatively analyze the difference in retinal thickness of the macular outer nuclear layer (ONL) between high myopia patients and normal subjects using spectral-domain optical coherence tomography (SD-OCT). Methods Seventy-eight participants (78 eyes) were divided into normal control group and high myopia group, according to the spherical equivalent (SE) of each eye. There were 24 cases (24 eyes) in the normal control group (-0. 25 D ~+0. 25 D) and 54 cases (54 eyes) in the high myopia group(SE<-6. 00 D). The macular retina was scanned using the posterior pole asymmetry analysis mode of the Heidelberg Spectralis (SD-OCT) system and divided into 9 areas according to Early Treatment of Diabetic Retinopathy Study. Quantitative analysis was performed to compare the difference in retinal thickness of the macular ONL in each area between the high myopia and normal control groups. Results The ONL thickness in the central area of the macular retina was significantly higher than that in the superior, nasal, inferior, and temporal areas (P < 0. 001 for all comparisons). The ONL thickness in the inferior area of the macular retina was significantly lower than that in the central, superior, nasal, and temporal areas (P < 0. 001 for all comparisons). In other sub-regions, no difference in the ONL thickness was found. The ONL thickness in the central, superior, nasal, inferior, and temporal areas was significantly lower in the high myopia group than in the normal control group (P < 0. 05 for all comparisons). Conclusion Regional variation is observed in the ONL thickness. The ONL thickness in the macular area of patients with high myopia is thinner.

ABSTRACT

Objective To evaluate the reproducibility of macular ganglion cell layer(GCL)measurements with high?resolution spectral domain?optical coherence tomography(SD?OCT)in both normal people and glaucoma patients. Methods In this study,24 normal subjects and 21 glauco?ma patients were prospectively included. Macular GCL thickness in 9 areas defined by Early Treatment Diabetic Retinopathy Study was measured with Spectralis SD?OCT applying posterior pole asymmetry analysis pattern. Within?subject standard deviation(Sw),coefficient of variation(CV) and intraclass correlation coefficient(ICC)in normal subjects and glaucoma patients were assessed. Results In normal subjects,the GCL thick?ness in macular central area was 12.58±2.69μm,the average GCL thickness in inner ring area was 48.87±3.81μm,the average GCL thickness in outer ring area was 37.28±1.75μm,and the GCL thickness mapping in normal subjects was horseshoe?shaped with opening to temporal. In glauco?ma patients,the GCL thickness in central area was 9.57±2.06μm,the average GCL thickness in inner ring area was 34.70±9.67μm,the average GCL thickness in outer ring area was 28.20±5.51μm,and the GCL thickness in every macular area in glaucoma eyes was thinner than that in normal eyes(P<0.001). For measurements of GCL thickness in normal subjects,Sw was 0.46 to 0.87μm,CV was 0.67%to 3.71%,and ICC was 0.904 to 0.977. For measurements of GCL thickness in glaucoma patients,Sw was 0.53 to 1.65μm,CV was 1.18%to 5.75%,and ICC was 0.833 to 0.993. Conclusion Spectralis SD?OCT had an excellent reproducibility for measurements of GCL thickness in both normal people and glaucoma patients, which is a reliable technique for evaluating longitudinal change and follow?up in glaucoma.

ABSTRACT

Objective to measure peripapillary atrophy(PPA)in glaucoma with high-resolution spectral domain-optical coherence tomography (SD-OCt)and conventional color fundus photograph,and compare the consistency of the results between the two imaging methods. Methods A to-tal of 30 patients(54 eyes)with glaucoma from Department of Opthalmology,the First Hospital,China Medical University were prospectively includ-ed for the study. SD-OCt images and color fundus photographs taken through the region of PPA were qualitatively analyzed. the average width of β-PPA area in 6 reference lines centered optic nerve were measured using SD-OCt and color fundus photograph. Correlation and agreement between the two measuring methods were analyzed. Results the average width of β-PPA was 229.89±99.82 μm using color fundus photographs and 224.14±97.10 μm using SD-OCt,respectively. there was no significant difference(P = 0.280). the correlation coefficient was 0.825 between re-sults measured by SD-OCt and color fundus photographs for β-PPA area(P < 0.001),and agreement between the two methods was good(ICC=0.923,95% CI 0.870-0.954). Conclusion the correlation and consistency is good between SD-OCt and conventional color fundus photograph measuring β-PPA area. β-PPA area can be estimated with SD-OCt.
